Effects of asparaginases and L-carnitine on Western-diet-induced hepatosteatosis in mice

Abstract Background: Asparaginases are common chemotherapeutic agents used for the treatment of acute lymphoblastic leukemia as a single or combinational therapy. Accompanying hepatotoxicity makes its use in elderly patients with pre-conditions, obesity other hepatopathies, difficult. Various hepatoprotective compounds like, L-carnitine, discussed to ameliorate induced hepatotoxicity. Methods: Here we aimed establish mouse model study effect asparaginases (L-asparaginase and Oncaspar) L-carnitine on Western-diet-induced hepatosteatosis mice. Dose-escalation studies were performed analyze C57BL/6 mice normal fatty livers. Subsequently, improve toxicity was tested. Results: Our results showed mild-to-moderate hepatotoxic effects while Western-diet higher degree vacuolization hepatocyte damage liver tissue. Testing established models did not show any protective impairment efficacy asparaginases. Conclusion: The here able demonstrate asparaginase-induced which enhanced by Western-diet. However, test potential ameliorating drugs, might need some improvements.
